You can get a penalty, if you have not delivered your job in time and you also can get one, when you deliver a damaged trailer. A third way to get a penalty is due to traffic offences, if you have enabled the "traffic offence" in your settings. (You also can disable this one) Also keep in mind that: Time in MP runs differently than on single player. It goes slower, but never stops. If you pick a job before actually connecting to the server, the time in your profile is way behind the server's one. So as soon as you start driving, you are already late.
